Peripheral Blood Stem Cells Harvesting via Apheresis in a Sheep Model Following G-CSF Stimulation

Abstract

This study aimed to investigate the ease of harvesting autologous peripheral blood stem cells (PBSC) in a sheep model via apheresis. Seven domestic sheep underwent daily subcutaneous injections of G-CSF (Neupogen; Amgen, Thousand Oaks, CA) for three days prior to the apheresis procedure. Blood samples were collected before G-CSF administrations. On the fourth day, autologous PBSC were collected using an automated cell separator during apheresis, and cryopreserved. White blood cell (WBC) count, viable cell counts, cell viability and PBSC population were determined and charted. Six out of the seven sheep successfully underwent the apheresis procedure. There were no adverse events found in all the sheep which received the G-CSF injections and underwent apheresis. Mean PBSC population in this study was 5.77% ± 2.72 where the lower WBC count tended to have lower PBSC population. In conclusion, it is possible to obtain PBSC via apheresis in sheep model following G-CSF stimulation.
